A Comparative Study On Microtuberization In Vitro And Colchicine Contents Of Two Species Of Gloriosa Of Colchicaceae. The regeneration potentiality and the process of microtuberization from apical buds of underground tubers of two species of Gloriosa were studied. The species included G. rothschildiana, a common hexaploid (2n=66) species of Gloriosa, and G. orangea, a rather uncommon tetraploid (2n=44) species. Both the species were propagated in vitro through bud multiplication using modified MS (Murashige and Skoog, 1962) basal media supplemented with 6-benzylaminopurine (BAP) and α-naphthaleneacetic acid (NAA) in presence of two different concentrations of sucrose (3 and 6 %). The optimum medium containing 3 % sucrose for microtuberization produced about 180-240 tubers within 6 months or little less. However, the medium containing 6 % sucrose, though initiated microtuberization 2 weeks earlier than that with 3 % sucrose, did not regenerate efficiently. The microtubers from 3 % sucrose-containing media showed almost 97% germination rate. The best result was obtained in a medium containing BAP (17.76 µM) and NAA (2.15 µM) with about 18-24 shoots and 14-21 microtubers / shoot. Colchicines was extracted from the microtubers of both the species and analyzed through HPLC for quantification. A 1.7 fold increase in colchicines content in G. rothschildiana and a 2.0 fold increase in G. orangea were observed as compared to in vivo tubers. The extracted colchicines, when treated with Allium sativum L and Mucuna pruriens L. root tip cells, showed better efficacy than standard colchicines from Sigma. In both these test plant systems, the extracted colchicines induced both polyploidy and diplochromatid formation at variable rates and the values were little higher in G. orangea than in G. rothschildiana. However, G. orangea responded better than G. rothschildiana against the same concentrations of plant growth regulators and showed better colchicines accumulation as well.
